Overview
Methionine synthase reductase (MTRR) is required for the reductive methylation of vitamin B12, which is essential for reactivating methionine synthase (MTR), and to maintain low circulating levels of plasma total homocysteine. A common mutation at position 66 within the MTR gene results in a transition from adenine to guanine. This MTRR A66G mutation has been shown to be significantly associated with maternal risk for neural tube defects and to predict response to selective serotonin reuptake inhibitor antidepressants in late life depression. In one study of treatment of resistant depressed patients the combination of MTRR A66G with MTR A2756G or MTHFR C677T polymorphisms resulted in a significantly greater response to L-methyltetrahydrofolate treatment.
